The seminar presented addressed aspects related to the progress of gene construction for the development of transgenesis in Rhodnius prolixus, a vector of Chagas disease. In which we discussed as a group the approach of transgenesis technology involving transposon for the production of insects expressing fluorescent reporter genes for membrane and nucleus. Thus, each region of the cloning vector was demonstrated expositorily. An important point of our group discussion was guided by the aspects involved in the optimization of the codons present in the coding region of the insert in the plasmid. Since there is still no bank of optimized codons for R. prolixus, this stage of the work was carried out by manual checking using as reference approximately 30 preferred codons of some proteins already described for this insect in the literature. This guarantees us a greater chance of transcription of the mRNA into protein in the gene construction. Finally, another important point presented to the group was the technical steps involved in the methodology of obtaining and injecting the DNA with the construction in the barber bug egg, where the main limitations and challenges of this stage were addressed.
