I presented a high molecular weight DNA extraction protocol for sequencing using Oxford Nanopore technology. The protocol features extraction of intact DNA, long fragments (N50=25Kb), a large amount of material, and elimination of short DNA fragments. The high throughput in library construction resulted in high flowcell occupancy and an output of approximately 100Gb. These results aid in understanding the technology and can be extended to other plant species.
